Would I need to learn Flash? I have Premiere CS3 and After Effects 7.0

You'd probably have to get a masters degree or doctorate in chemical engineering or perhaps biotechnology. Tethered Vesticles are nothing an amateur should be messing with. Besides, they sound slightly naughty.
Seriously... It's possible there is a third party application that creates this kind of presentation in flash. If not, you would have to learn flash. I've been struggling with it a lot lately. It seems it will do 95% of what I need, but I can't get past that last 5% and that ends up killing the project.
If you have a client that wants this kind of presentation and you have no experience in flash, you could always contract that work out.

Actually, it looks like they're using TechSmith ExpressShow, which is part of Camtasia Studio.
